凈化工(gong)作(zuo)(zuo)(zuo)臺(tai)是一(yi)種局部(bu)層流裝置,能(neng)在局部(bu)形成(cheng)高(gao)潔(jie)度的工(gong)作(zuo)(zuo)(zuo)環境(jing)。它由工(gong)作(zuo)(zuo)(zuo)臺(tai)、過(guo)(guo)濾(lv)器(qi)、風機、靜(jing)壓(ya)箱(xiang)和支(zhi)撐體等組成(cheng),采用(yong)過(guo)(guo)濾(lv)空(kong)(kong)(kong)氣使(shi)工(gong)作(zuo)(zuo)(zuo)臺(tai)操(cao)作(zuo)(zuo)(zuo)區達到凈化除(chu)(chu)菌(jun)的目的。室內空(kong)(kong)(kong)氣經(jing)預過(guo)(guo)濾(lv)器(qi)和高(gao)效過(guo)(guo)濾(lv)除(chu)(chu)塵后(hou)以垂直或水平層流狀(zhuang)態(tai)通過(guo)(guo)工(gong)作(zuo)(zuo)(zuo)臺(tai)的操(cao)作(zuo)(zuo)(zuo)區,由于空(kong)(kong)(kong)氣沒有(you)渦流,所以,任(ren)何一(yi)點灰(hui)塵或附(fu)著(zhu)在灰(hui)塵上的雜菌(jun)都(dou)能(neng)被排除(chu)(chu),不易向別處(chu)擴散和轉(zhuan)移。因此,可使(shi)操(cao)作(zuo)(zuo)(zuo)區保持無菌(jun)狀(zhuang)態(tai)。
與沒有細菌檢測室和注射箱相當,選用濾出操作步驟臺兼有操作步驟經濟條件好、操作步驟利于、沒有細菌檢測功能好是真的嗎、無空氣消毒藥物制劑對人損害、負載動用面積小且可走動等的優勢。假若放至沒有細菌檢測內選用,沒有細菌檢測功能好效果更好。其優點是價位價格較高,預濾出器和效率濾出器還須要限期沖洗和調換。 油田蒸氣高壓蒸汽滅菌鍋高(gao)(gao)壓蒸(zheng)(zheng)汽(qi)(qi)(qi)滅菌(jun)(jun)鍋(guo)(guo)是一(yi)個密閉的、可(ke)以耐受一(yi)定(ding)壓力(li)的雙層金屬鍋(guo)(guo)。鍋(guo)(guo)底或(huo)夾層內(nei)盛水,當水在鍋(guo)(guo)內(nei)沸騰時(shi)由于蒸(zheng)(zheng)汽(qi)(qi)(qi)不能(neng)逸出(chu),使鍋(guo)(guo)內(nei)壓力(li)逐(zhu)漸升(sheng)高(gao)(gao),水的沸點和溫度可(ke)隨之升(sheng)高(gao)(gao),從(cong)而達到(dao)高(gao)(gao)溫滅菌(jun)(jun)的目的。一(yi)般(ban)在0.11MPa的壓力(li)下,121℃滅菌(jun)(jun)20~30min,包括芽孢在內(nei)的所有微生物(wu)均可(ke)被(bei)殺死。如果滅菌(jun)(jun)物(wu)品體積較大,蒸(zheng)(zheng)汽(qi)(qi)(qi)穿透困難,可(ke)以適當提高(gao)(gao)蒸(zheng)(zheng)汽(qi)(qi)(qi)壓力(li)或(huo)延長(chang)滅菌(jun)(jun)時(shi)間(jian)。
超直流電殺菌鍋有離心式、直列V型、手堤式等很多的種方式,在微微生物發酵學實驗所室,極為實用的是手堤式和直列V型超直流電水汽殺菌鍋。和過熱蒸汽殺菌鍋想必,超直流電殺菌鍋的特點是殺菌想要的時段短、節省助燃劑、殺菌完全等。其優點是價格多少珍貴,殺菌容積較小。 陪養箱培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang)是(shi)培(pei)養(yang)(yang)(yang)(yang)微生物(wu)的專用設備(bei)。制熱式培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang)是(shi)由(you)電(dian)爐絲(si)和溫(wen)度控(kong)制儀合成的固(gu)定體積(ji)的恒(heng)溫(wen)培(pei)養(yang)(yang)(yang)(yang)裝(zhuang)置,大小規格不(bu)一(yi)。微生物(wu)實驗室常用的培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang)工(gong)作容積(ji)有450×450×350mm3或650×500×500mm3,適用于室溫(wen)至60℃之間的各類(lei)微生物(wu)培(pei)養(yang)(yang)(yang)(yang)。目前(qian),隨著(zhu)科學水平的發展,培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang)設備(bei)的完(wan)善程度和價格有很大差別(bie)。有各種結(jie)構合理、功(gong)能齊全的培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang),如恒(heng)溫(wen)培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang)、恒(heng)溫(wen)恒(heng)濕(shi)培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang)、低(di)溫(wen)培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang)、微生物(wu)多用培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang)和二氧化碳培(pei)養(yang)(yang)(yang)(yang)箱(xiang)(xiang)(xiang)等。有的用計算機控(kong)制,可選擇多條時間線變換溫(wen)差,從而(er)克服了環境溫(wen)度的影響(xiang),一(yi)年四(si)季均能達到培(pei)養(yang)(yang)(yang)(yang)要(yao)求(qiu)的溫(wen)度。
微生物多用培養箱是(shi)集(ji)加(jia)熱、制(zhi)冷和振蕩于一(yi)體(ti)(ti)的微生物液(ye)體(ti)(ti)發酵裝置。工作室的溫度在(zai)15~50℃范圍(wei)內任意選(xuan)定(ding),選(xuan)定(ding)后經(jing)溫控(kong)(kong)儀自(zi)動控(kong)(kong)制(zhi),保(bao)持工作室內恒溫。同時設有(you)可控(kong)(kong)硅調速(su)系(xi)統,振蕩機(ji)轉速(su)可在(zai)1~220rpm范圍(wei)內任意調控(kong)(kong)。
非常烘干箱干(gan)(gan)燥箱是(shi)用于除(chu)去(qu)潮濕物(wu)料內及(ji)器皿內外水分(fen)或(huo)其(qi)它揮發(fa)性溶液的設備。類型很多,有箱式(shi)(shi)、滾筒式(shi)(shi)、套間式(shi)(shi)、回(hui)轉(zhuan)式(shi)(shi)等。微生物(wu)學實驗室(shi)多用箱式(shi)(shi)干(gan)(gan)燥箱,大小規格不一。工作室(shi)內配(pei)有可活動(dong)的鐵絲網板,便于放置被(bei)干(gan)(gan)燥的物(wu)品。制(zhi)(zhi)熱(re)升溫(wen)(wen)式(shi)(shi)干(gan)(gan)燥箱也是(shi)有電爐絲和(he)溫(wen)(wen)度(du)控(kong)制(zhi)(zhi)儀組(zu)成(cheng),可調節溫(wen)(wen)度(du)從室(shi)溫(wen)(wen)至(zhi)300℃任意選(xuan)擇。有的干(gan)(gan)燥箱采用導電溫(wen)(wen)度(du)計為敏感元件,配(pei)合晶體管和(he)繼電器組(zu)成(cheng)自動(dong)控(kong)制(zhi)(zhi)系統,克服了金屬管型熱(re)膨脹控(kong)制(zhi)(zhi)的缺點。此外,還有真空(kong)(kong)干(gan)(gan)燥箱(配(pei)有真空(kong)(kong)泵和(he)氣(qi)壓表),可在常壓或(huo)減壓下操作。
搖床
搖(yao)(yao)(yao)床(chuang)又稱(cheng)搖(yao)(yao)(yao)瓶(ping)機,它是培(pei)養(yang)好(hao)氣(qi)性微生物的小型(xing)試驗設備或(huo)作為(wei)種子(zi)擴大培(pei)養(yang)之(zhi)用(yong),常用(yong)的搖(yao)(yao)(yao)床(chuang)有往(wang)復(fu)式(shi)和旋(xuan)轉式(shi)兩種。往(wang)復(fu)式(shi)搖(yao)(yao)(yao)床(chuang)的往(wang)復(fu)頻率一(yi)般在(zai)80~140次/min,沖程一(yi)般為(wei)5~14cm,如頻率過(guo)快、沖程過(guo)大或(huo)瓶(ping)內液體裝量過(guo)多,在(zai)搖(yao)(yao)(yao)動時液體會濺到包(bao)扎瓶(ping)口(kou)的紗布或(huo)棉塞(sai)上,導致雜(za)菌污染,特別是啟(qi)動時更容易發(fa)生這種情況。旋(xuan)轉式(shi)搖(yao)(yao)(yao)床(chuang)的偏心距一(yi)般在(zai)3~6cm之(zhi)間,旋(xuan)轉次數為(wei)60~300rpm。
放(fang)在搖床(chuang)(chuang)上的(de)(de)培(pei)養瓶(ping)(ping)(一般為三角瓶(ping)(ping))中的(de)(de)發酵液所(suo)需要的(de)(de)氧是由空氣(qi)經瓶(ping)(ping)口(kou)包扎的(de)(de)紗(sha)布(一般8層)或棉(mian)塞通入的(de)(de),所(suo)以氧的(de)(de)傳遞與瓶(ping)(ping)口(kou)的(de)(de)大(da)小、瓶(ping)(ping)口(kou)的(de)(de)幾(ji)何形(xing)狀、棉(mian)塞或紗(sha)布的(de)(de)厚(hou)度和密(mi)度有關。在通常(chang)情況下,搖瓶(ping)(ping)的(de)(de)氧吸(xi)收系數取決于搖床(chuang)(chuang)的(de)(de)特性(xing)和三角瓶(ping)(ping)的(de)(de)裝樣量。

注射箱接(jie)種箱(xiang)(xiang)(xiang)分為(wei)固(gu)體(ti)(ti)菌(jun)種接(jie)種箱(xiang)(xiang)(xiang)和液體(ti)(ti)菌(jun)種接(jie)種箱(xiang)(xiang)(xiang)兩(liang)種。固(gu)體(ti)(ti)菌(jun)種接(jie)種箱(xiang)(xiang)(xiang)是(shi)一個(ge)用(yong)木(mu)料和玻(bo)(bo)璃制(zhi)成(cheng)或由有機玻(bo)(bo)璃焊接(jie)而成(cheng)的(de)(de)(de)密(mi)閉小(xiao)箱(xiang)(xiang)(xiang)。又分為(wei)雙(shuang)人(ren)(ren)和單(dan)人(ren)(ren)操(cao)作(zuo)箱(xiang)(xiang)(xiang)。箱(xiang)(xiang)(xiang)體(ti)(ti)可(ke)大可(ke)小(xiao),一般箱(xiang)(xiang)(xiang)體(ti)(ti)長約143cm,寬86cm,總高154cm,支架76cm。箱(xiang)(xiang)(xiang)的(de)(de)(de)上(shang)部左右兩(liang)側各裝(zhuang)有兩(liang)扇能啟閉的(de)(de)(de)玻(bo)(bo)璃推拉門,方便菌(jun)種進(jin)出(chu)。窗的(de)(de)(de)下部分別設(she)有兩(liang)個(ge)直徑約13cm的(de)(de)(de)圓洞(dong),兩(liang)洞(dong)的(de)(de)(de)中(zhong)心距離為(wei)52cm(同肩寬),洞(dong)口裝(zhuang)有帶松緊帶的(de)(de)(de)袖(xiu)套(tao),以防雙(shuang)手(shou)在箱(xiang)(xiang)(xiang)內操(cao)作(zuo)時,外(wai)界空(kong)氣進(jin)入箱(xiang)(xiang)(xiang)內造成(cheng)污染(ran)。操(cao)作(zuo)時兩(liang)人(ren)(ren)相(xiang)對而坐,雙(shuang)手(shou)通(tong)過袖(xiu)套(tao)伸入箱(xiang)(xiang)(xiang)內。箱(xiang)(xiang)(xiang)兩(liang)側最(zui)好也(ye)裝(zhuang)上(shang)玻(bo)(bo)璃,箱(xiang)(xiang)(xiang)頂(ding)部為(wei)木(mu)板(ban)或玻(bo)(bo)璃。箱(xiang)(xiang)(xiang)內頂(ding)部裝(zhuang)有紫外(wai)線殺(sha)菌(jun)燈和照明(ming)用(yong)日光燈各一支。箱(xiang)(xiang)(xiang)體(ti)(ti)安裝(zhuang)木(mu)板(ban)或玻(bo)(bo)璃均可(ke),但要注(zhu)意密(mi)封(feng)。
液體(ti)(ti)菌(jun)(jun)種(zhong)接(jie)種(zhong)箱(xiang)是(shi)專(zhuan)為移接(jie)液體(ti)(ti)菌(jun)(jun)種(zhong)而(er)設計(ji)的。比固體(ti)(ti)菌(jun)(jun)種(zhong)箱(xiang)窄長(chang),單側兩人(ren)操(cao)作(zuo)。內設軌道和(he)紫外線燈(deng),箱(xiang)兩端開(kai)有(you)高25cm,寬10cm的長(chang)方形(xing)出口(kou),方便菌(jun)(jun)種(zhong)進出,洞(dong)口(kou)設有(you)小(xiao)推門。進出口(kou)下(xia)處設蒸汽源,接(jie)種(zhong)時用(yong)蒸汽封住進出口(kou),以防(fang)雜菌(jun)(jun)進入箱(xiang)內。箱(xiang)背面設有(you)液體(ti)(ti)菌(jun)(jun)種(zhong)移接(jie)管能進入的小(xiao)孔。
接(jie)(jie)種(zhong)箱滅菌(jun)時(shi),用(yong)紫(zi)外(wai)線(xian)照(zhao)射30min。如果沒有(you)紫(zi)外(wai)線(xian)燈(deng),可(ke)用(yong)甲(jia)醛和(he)高(gao)錳(meng)酸鉀(甲(jia)醛10~14mL/m3+高(gao)錳(meng)酸鉀5~7g/m3空(kong)間)熏(xun)蒸(zheng)30min以上。使(shi)用(yong)時(shi),先將所需物品(pin)和(he)工具放入接(jie)(jie)種(zhong)箱內,然(ran)后進行藥(yao)劑(ji)熏(xun)蒸(zheng)和(he)紫(zi)外(wai)線(xian)滅菌(jun),再按無菌(jun)操作(zuo)進行接(jie)(jie)種(zhong)。接(jie)(jie)種(zhong)箱的結構簡單,造價低廉,易消(xiao)毒滅菌(jun),操作(zuo)方便,而(er)且(qie)人在箱外(wai)操作(zuo),氣溫較高(gao)時(shi)也能作(zuo)業。缺點是進出培養基費工費時(shi),每(mei)次(ci)接(jie)(jie)種(zhong)前都需要進行滅菌(jun)。
冰箱冷藏 微生物實驗室的冰箱主要有兩種:普通冰箱和低溫冷凍冰箱。普通冰箱一般都具有兩個柜子,即鮮藏柜和冷藏柜,溫度分別為4℃和-20℃;低溫冷凍冰箱溫度一般控制在-40~-80℃。它們都可以用于微生物菌種保藏。鮮藏柜常用于保存斜面菌種,保藏時間在3個月左右。超過3個月,斜面就會變干,因此需要轉接菌種。如果要長時間保存菌種,則需要經過處理后,貯藏于普通冰箱的冷藏柜或低溫冷凍冰箱中,它們的保藏時間較長,一般都在1年以上。
